Dosage form
50 mg powder/vial
Dose
Adult : Initial dose: 0.25 mg/kg. Increase by daily increments of 0.25 mg/kg or faster if tolerated.
Maximum of 1.5 mg/kg/24 h.
Administration
IV: Intermittent infusion
Intravenous Concentration
Intermittent Infusion: 0.1 mg per mL.
Up to 0.25 mg per mL via central venous access only
Intravenous Rate
Intermittent Infusion: Infuse over 2 to 6 hours

Adverse effect
Febrile reactions (fever, chills), headache, anorexia, vomiting, malaise, nephrotoxicity, hypokalaemia, hypomagnesaemia, cardiac dysrhythmias, neurological disorders, blood disorders, liver dysfunction, pain at the injection site, thrombophlebitis, anaphylactoid reactions, rash
Precaution
Check for hypersensitivity to amphotericin before administration. Use with caution in patients with renal and/or hepatic failure. Monitor electrolytes and fluid balance. Avoid extravasation, peripheral venous infusion of concentrated solutions, or, rapid infusion in patients with renal failure. Avoid concurrent granulocyte infusions due to the adverse pulmonary effects
Contraindication
Information
Protection from light is usually unnecessary if infused within 6 hours of preparation – avoid direct sunlight. Heparin 1 unit per 1mL has been added to dilute infusion solutions to reduce the incidence of phlebitis and pain on peripheral intravenous injection. Give an antipyretic prior to the infusion
